Comparative_Evaluation_of_the_Salivary_and_Buccal_Mucosal_Microbiota_by_16S_rRNA_Sequencing_for_Forensic_Investigations

Publication: The_human_microbiome_has_emerged_as_a_new_potential_biomarker_for_forensic_investigations_with_the_development_of_high-throughput_sequencing_and_bioinformatic_analysis_during_the_last_decade._The_oral_cavity_has_many_different_microbial_habitats,_with_each_habit_colonized_by_specific_and_individualized_microbiota._As_saliva_and_buccal_mucosa_are_common_biological_evidence_in_forensic_science,_understanding_the_differences_of_microbial_communities_between_the_two_is_important_for_forensic_original_identification._Moreover,_the_oral_microbiota_is_individualized,_whereas_there_are_few_studies_on_the_application_of_forensic_personal_identification_that_need_to_be_supplemented._In_this_study,_Streptococcus_was_the_most_abundant_genus,_with_an_average_relative_abundance_of_49.61%_in_the_buccal_mucosa,_while_in_the_saliva,_Streptococcus,_Veillonella,_and_Neisseria_had_similar_proportions_(20%,_15%,_16%)_and_were_the_dominant_genera._The_¦Á_and_¦Â_diversity_displayed_a_significant_distinctness_between_the_saliva_and_buccal_mucosal_groups._The_community_assembly_mechanism_stated_that_the_deterministic_process_played_a_more_significant_effect_in_shaping_the_salivary_bacterial_community_assembly_than_buccal_mucosa,_which_explained_the_microbial_differences._Of_the_test_samples,_93.3%_can_be_correctly_classified_with_the_random_forest_model_based_on_the_microbial_differences._Targeting_the_low-abundance_bacteria_at_the_species_level,_52%_of_experimental_participants_could_be_discriminated_by_using_the_observed_unique_bacterial_species._In_conclusion,_the_salivary_bacterial_community_composition_differed_from_that_of_the_buccal_mucosa_and_showed_high_richness_and_diversity._With_the_random_forest_model,_the_microbiota_of_saliva_and_buccal_mucosa_can_be_classified,_which_can_be_used_in_identifying_the_source_of_oral_biological_trace._Furthermore,_each_individual_has_a_unique_bacterial_community_pattern,_and_the_presence_or_absence_of_unique_bacteria_and_differences_in_the_composition_of_the_core_oral_microbiota_are_the_key_points_for_forensic_personal_discrimination_that_supplement_the_study_of_oral_microbial_application_to_forensic_personal_discrimination._Whether_for_original_identification_or_personal_discrimination,_the_oral_microbiome_has_great_potential_for_application.
