Metataxonomic_and_metabolomic_evidence_of_biofilm_homeostasis_disruption_related_to_caries:_An_in_vitro_study

Publication: The_ecological_dysbiosis_of_a_biofilm_includes_not_only_bacterial_changes_but_also_changes_in_their_metabolism._Related_to_oral_biofilms,_changes_in_metabolic_activity_are_crucial_endpoint,_linked_directly_to_the_pathogenicity_of_oral_diseases._Despite_the_advances_in_caries_research,_detailed_microbial_and_metabolomic_etiology_is_yet_to_be_fully_clarified._T_o_advance_this_knowledge,_a_meta-taxonomic_approach_based_on_16S_rRNA_gene_sequencing_and_an_untargeted_metabolomic_approach_based_on_an_ultra-high_performance_liquid_chromatography-quadrupole_time-of-flight_mass_spectrometry_analysis_(UHPLC/Q-TOF-MS)_were_conducted._T_o_this_end,_an_in_vitro_biofilm_model_derived_from_the_saliva_of_healthy_participants_were_developed,_under_commensal_and_cariogenic_conditions_by_adding_sucrose_as_the_disease_trigger._The_cariogenic_biofilms_showed_a_significant_increase_of_Firmicutes_phyla_(p_=_0.019),_due_to_the_significant_increase_in_the_genus_Streptococcus_(p_=_0.010),_and_Fusobacter_(p_<_0.001),_by_increase_Fusobacterium_(p_<_0.001)_and_Sphingomonas_(p_=_0.024),_while_suffered_a_decrease_in_Actinobacteria_(p_<_0.001)._As_a_consequence_of_the_shift_in_microbiota_composition,_significant_extracellular_metabolomics_changes_were_detected,_showed_59_metabolites_of_the_120_identified_significantly_different_in_terms_of_relative_abundance_between_the_cariogenic/commensal_biofilms_(Rate_of_change_>_2_and_FDR_<_0.05)._Forty-two_metabolites_were_significantly_higher_in_abundance_in_the_cariogenic_biofilms,_whereas_17_metabolites_were_associated_significantly_with_the_commensal_biofilms,_principally_related_protein_metabolism,_with_peptides_and_amino_acids_as_protagonists,_latter_represented_by_histidine,_arginine,_L-methionine,_glutamic_acid,_and_phenylalanine_derivatives.
