Polymicrobial_Aggregates_in_Human_Saliva_Build_the_Oral_Biofilm

Publication: Biofilm_community_development_has_been_established_as_a_sequential_process_starting_from_the_attachment_of_single_cells_on_a_surface._However,microorganisms_are_often_found_as_aggregates_in_the_environment_and_in_biological_fluids._Here,we_conduct_a_comprehensive_analysis_of_the_native_structure_and_composition_of_aggregated_microbial_assemblages_in_human_saliva_and_investigate_their_spatiotemporal_attachment_and_biofilm_community_development._Using_multiscale_imaging,cell_sorting,and_computational_approaches_combined_with_sequencing_analysis,a_diverse_mixture_of_aggregates_varying_in_size,structure,and_microbial_composition,including_bacteria_associated_with_host_epithelial_cells,can_be_found_in_saliva_in_addition_to_a_few_single-cell_forms._Phylogenetic_analysis_reveals_a_mixture_of_complex_consortia_of_aerobes_and_anaerobes_in_which_bacteria_traditionally_considered_early_and_late_colonizers_are_found_mixed_together._When_individually_tracked_during_colonization_and_biofilm_initiation,aggregates_rapidly_proliferate_and_expand_tridimensionally,modulating_population_growth,spatial_organization,and_community_scaffolding._In_contrast,most_single_cells_remain_static_or_are_incorporated_by_actively_growing_aggregates._These_results_suggest_an_alternative_biofilm_development_process_whereby_aggregates_containing_different_species_or_associated_with_human_cells_collectively_adhere_to_the_surface_as_¡°growth_nuclei¡±_to_build_the_biofilm_and_shape_polymicrobial_communities_at_various_spatial_and_taxonomic_scales.
